猪血浆中凝血酶、免疫球蛋白等功能蛋白的联合提取

摘要: 实验对猪血浆中白蛋白、免疫球蛋白、纤维蛋白原及凝血酶等功能蛋白进行了联合提取。通过对各步骤中蛋白的检测确立了一套有效的联合提取方法:将柠檬酸钠抗凝的血浆经冻融处理,离心得到纤维蛋白原沉淀,上清通过吸附法提取凝血酶后再经盐析法分离提取白蛋白及免疫球蛋白。1L血浆可同时提得白蛋白31.41g、免疫球蛋白13.86g、凝血酶粗品0.22g(比活力为38.5U/mg)、纤维蛋白原为2.67g。

Abstract: A effective method of extracting functionality protein such as albumin、immunoglobulin、fibrinogen and thrombin from pig plasma was introduced in this article. Extracting fibrinogen with freeze-thawing method and thrombin from supernatant by absorbing test, then salt out to distill albumin and immunoglobulin. In 1L pig plasma ,about 31.41g albumin and 13.86g immunoglobulin and 0.22g thrombin and 2.67g fibrinogen have been extracted. The specific activity of thrombin is 38.5U/mg.
