Studies on Interaction between Protein A and Immunoglobul in G Using Real-Time BIA Technique

Abstract

Abstract:

The interaction betw een protein A and MIgG has been investigated.Some interesting kinetic parameters have been obtained.The kinetic constants between protein A and MIgG that we have obtained f rom the BIA senso rg ram is as follow ing :associate rate constant :ka=5.07×10⁴ (mol/L)^-1s^-1, dissociate rate constant :kd=9.65×10^-5(s^-1), af finity constant :KA=5.25×10⁸(mol/L)^-1.The concentrat ion detection of MIgG, using protein A immobilized sensor chip, has been also studied.A good linear relationship betw een SPR response unit s and MIgG concent ration has been found in the range of 0.64 ～ 12mg/ L.
